On the drawer base cabinet, if the drawer height is 12 inches and select the 4 inch tall standard toe kick will the total height of the unit be 16 inches (drawer height + toe kick height)?

Sure, ordering a 12” HEIGHT base cabinet will yield a 12” tall cabinet that has a 4” tall toe kick included in that height. Toe kicks are never added to the HEIGHT dimension:
